Spider-Man's conflict with Doctor Strange in Spider-Man: No Way Home is exactly what he needs to grow. In addition to facing the legion of villains from the multiverse, it seems that the wall-crawler is set to take on the Sorcerer Supreme as well. Ironically, this sort of antagonism may actually serve the character rather than harm him.
As the trailers show, No Way Home will focus on the fallout from Spider-Man tampering with the spell Strange casts in order to erase everyone knowing he's Peter Parker. This leads to several villains from the multiverse being released, forcing him to take them all on. However, the second trailer throws in a new twist that further complicates matters. After discovering that the villains will be transported back to die in their universes, he tries to stop Strange from sending them back, which leads to the two going against each other.
While undoubtedly challenging, Spider-Man's conflict with Doctor Strange in No Way Home is what he needs to truly grow and come into his own. While it's undeniable that the MCU's Spider-Man has proven to be a true hero in the past, it's also apparent that he's been far too reliant on other heroes to help him on his journey, particularly Iron Man. Going against Strange to do what he believes is right will change this.
Spider-Man going against Doctor Strange shows that he is now breaking away from relying on other heroes to help him do the right thing. In both Spider-Man: Homecoming and Spider-Man: Far From Home, a large part of his arc involves him trying to live up to Iron Man's legacy. This leads to both films leaning heavily on Tony Stark's father-like involvement in Spider-Man's life. While this helps build the bond between the two, it also takes away from Spider-Man doing heroic things apart from his mentor. The dispute with Strange proves that he is now grown to a place where he can be heroic on his own without the need for another super-powered ally.
Spider-Man's feud with Doctor Strange will also help him truly understand what sacrifices will need to be made as a hero. Since Spider-Man is trying to save the villains from dying in their universes, Strange will be able to teach him through their conflict that as a hero, he cannot save everyone. Given his knowledge of the multiverse, Strange understands keeping No Way Home's villains alive will have disastrous consequences. As the second trailer shows, Spider-Man seems to realize this too, as he declares that he can't save everyone. In that sense, his conflict with Strange may be what finally helps him truly realize that "with great power, comes great responsibility."
Of course, both Spider-Man and Doctor Strange will likely put aside their differences to work together in the finale of Spider-Man: No Way Home. However, this disagreement must hold more weight than a mere skirmish thrown in to briefly shake things up. With a new trilogy on the horizon, it should ideally be what brings Spider-Man's current MCU arc to a satisfying close, as it molds the wall-crawler into a hero who has grown to a point where he is ready to take on challenges by himself.
